Delta delta Ct comparison & wild-type

Postby Henrik » Fri May 22, 2009 11:26 am

I haven't been able to figure out how to do a delta delta Ct comparison with my data to get the relative expression of genes in one sample versus another sample. Also I was wondering if you can combine data from multiple experiments - for example if you ran wild-type on one plate and mutant on another plate could you compare the expression of the mutant genes to the wild type

Dear Henrik,

Performing data pre-processing in GenEx's Data Editor will enable you to obtain delta delta Ct measures of gene expression by performing Normalization with reference genes and Relative quantities (and possibly other pre-processing steps as well).

In the same pre-processing menu you have functionality to perform interplate calibration which is a good way to combine data from multiple experiments.

The tutorial at http://www.multid.se/demonstration.html (click "Comparison of two groups in a nested design") explains all these steps in a comprehensive way.

Best wishes,
Anders
