calculation of ddCt wihout control group

Postby gentostat27 » Tue Jun 08, 2010 4:18 pm

Hey all

I am new at analysis of qPCR data and I have one question which I am
unable to find an answer to

- I have qpcr results for several genes and one housekeeping gene

- In contrast to grouping I would like to use a clinical variable which
is continuous and hence would like to take the qpcr value for each person
and put in into my regression equation (the same one i used for the
microarray data whose results i want to validate using qPCR)

- Since I do not have a control group nor a calibrator, I cannot use the
delta delta Ct method.

- would it be wrong if i used 2^-delta Ct value for each individual in my
regression equation?
(delta Ct = Ct of gene - Ct of housekeeper gene)

Thanks
